Ph-Negative Chronic Myeloid Leukemia: Molecular Analysis of ABL Insertion Into M-BCR on Chromosome

چکیده
Leukemic cells f rom a patient with Ph-negative chronic myeloid leukemia (CML) had a normal karyotype. M-BCR was rearranged and chromosome in si tu hybridization showed a n ABL insertion be tween 5’ and 3’ M-BCR on a n apparently normal chromosome 22. T h e association of 5’ BCR and 3’ ABL at t h e 5’ junction of t h e chromosome 9 insert was typical of t h a t found for t h e BCR-ABL fusion g e n e in o the r patients with t h e standard t(9;22) and CML. With a n M-bcr-3’ probe, we cloned and characterized a 3’ junction fragment. Field inversion gel electrophoresis and chromosome in si tu hybridization studies using a probe isolated from genomic DNA 5’ of t h e junction showed tha t

Frequency of BCR-ABL Fusion Transcripts in Iranian Azeri Turkish patients with Chronic Myeloid Leukemia
Background: The Philadelphia chromosome (Ph) characterized by t (9; 22) (q34; q11.2) is a reciprocal translocation giving rise to a chimeric BCR-ABL fusion gene. Incidence of Ph chromosome is over 98% in Patients with Chronic Myeloid Leukemia (CML) and around 20% in acute lymphoblastic leukemia (ALL). The finding of this fusion gene is essential for diagnosis of CML by detection of various fusi...
متن کاملPh-negative chronic myeloid leukemia: molecular analysis of ABL insertion into M-BCR on chromosome 22.
Leukemic cells from a patient with Ph-negative chronic myeloid leukemia (CML) had a normal karyotype. M-BCR was rearranged and chromosome in situ hybridization showed an ABL insertion between 5' and 3' M-BCR on an apparently normal chromosome 22. The association of 5' BCR and 3' ABL at the 5' junction of the chromosome 9 insert was typical of that found for the BCR-ABL fusion gene in other pati...
متن کاملDetection of abl/bcr Fusion Gene in Patients Affected by Chronic Myeloid Leukaemia by Dual-Colour Interphase Fluorescence in situ Hybridisation
Conventional cytogenetic is the standard technique for detection of Philadelphia (Ph) chromosome in chronic myeloid leukemia (CML). Evaluation of abelson murine leukemia/breakpoint cluster region (abl/bcr) fusion using dual-colour fluorescence in situ hybridization (D-FISH) is an alternative approach allowing rapid and reliable detection of the disease. We employed the technique of interphase D...
متن کاملAnalysis of Philadelphia chromosome-negative BCR-ABL-positive chronic myelogenous leukemia by hypermetaphase fluorescence in situ hybridization.
BACKGROUND In 5%-10% of patients with of chronic myelogenous leukemia (CML), the Philadelphia chromosome (Ph) is not identified, despite the presence of the associated BCR-ABL molecular abnormality (Ph-negative, BCR-ABL-positive CML) because of sub-microscopic rearrangements. PATIENTS AND METHODS Six patients with Ph-negative, BCR-ABL-positive CML were investigated. The Ph chromosome detectio...
متن کاملBCR-ABL fusion genes and laboratory findings in patients with chronic myeloid leukemia in northeast Iran
Background: A specific chromosomal abnormality, the Philadelphia chromosome (BCR-ABL fusion), is present in all patients with chronic myeloid leukemia (CML). The b2a2 and b3a2 fusion mRNAs encode p210 fusion protein p210 and e1a2 encode p190. The aim of this study was to evaluate the frequency of BCR-ABL fusion transcript variants in Northeast of Iranian CML patients and to compare the laborato...
